Overview

EZ-Bath™ Tissue Organ Baths, are high-performance, user friendly multi-channel tissue/organ pharmacology systems. The state-of-the-art organ bath is designed, based on many years of combined experiences of various scientists. The EZ-Bath™ have been used for pharmacological and preclinical studies of isolated vascular or non-vascular tissues, skeletal or smooth muscles, cardiac tissues/organs (e.g. atria, papillary, sinus node), and tissues from respiratory/lung, urinary tract, kidney, and intestine or colon, etc.

The EZ-Bath™ features multiple innovative, proprietary functions proven to ease tissue organ studies in all phases of experiments: tissue mounting, oxygenation control, preset tension, (stimulation), recording and data marking, waste disposal/washing, & data analyses. These functions allow more efficient experiments and producing of more and consistent isometric or isotonic data with sensitivity up to 0.2-0.5mg, across channels and experiments, in comparison to most others’ systems. In addition, the EZ-Baths systems are much more affordable and cost $3000 to $8000 less than others.

New functions include (1). embedded bio-sensing (nitric oxide, oxygen, glucose, epinephrine, pH) and/or bio-potential recording; (2). automated (optional) or manual (one-push valve) buffer filling, emptying and washing; (3). new tissue holders with built-in ring, spiral or zigzag Pt. stimulation electrodes. (4). upgrades, when appropriate, to EZ-NeWMyo™ Micro Vessel Needle and Wire Myograph or EZ-Fusion™ Tissue Superfusion System.

Applications

EZ-Bath have been widely used for pharmacology or toxicology study of tissue reactivity, morphology, and length-tension relationship, nitric oxide roles in tissue functions, etc. in following isolated biological tissues:

  • Isolated vascular rings: smooth muscle or endothelium cells;
  • - e.g., vascular muscle tone in relation to vasodilators or vasoconstrictors (acetylcholine, adenosine, epinephrine, etc.)

  • Isolated smooth/skeletal tissues or organs:
- skeletal muscles: e.g., roles of nitric oxide in skeletal muscle repair
- cardiac (atria, papillary, sinus node): e.g. study of heart muscle tone & nitric oxide-mediated tissue injury.
- adipose tissue
- bronchi, respiratory tract tissue
- urinary tract tissues
- intestine and colon tissues, etc.

Nitric oxide has been implicated in the functions of many biological tissues: adipose, skeletal muscle repair (L.I. Filippin, etc., Nitric Oxide, Vol 21, p 157-163, 2009), and vasopressure of blood vessels, etc. Our EZ-Bath-x-SED systems with bio-sensing option allows real time recording of extracellular nitric oxide concentration, an important tool to study these bio-functions.

    Tension and Force Transducers (included)

    i-FOT Series Isometric Force Transducers with max sensitivity of 0.2-0.5mg and range from 0-10 to 0-2000grams. The new SW series have both high sensitivity and wide dynamic range (200,000:1). They’re compatible with our BRAM-4B, i-BRIDGE transducers amplifiers, or BRAMDA-400/800 and i-DAQ 4/8 data acquisition systems. They can also be easily adapted to ADI and other suppliers’ systems. In addition, we also supply i-TFOT Isotonic Force Transducers, fiber-optic based force transducers, mini pressure, temperature sensors, etc.

    2. Bio-sensing, Direct/Field Tissue Stimulation and/or Extracellular Field Potential Recording:

    GlobalTown supplies a series of miniature biosensors that are used seamlessly for simultaneously measuring nitric oxide (NO), dissolved oxygen, glucose, epinephrine, norepinephrine, or bio-potential, etc., near tissue vessel rings under tension, drug perfusion and/or stimulation. This allows studies of interactions between tissue muscle tone and biochemical release or perfusion, e.g. effect of NO-mediated heart tissue injury on muscle tone, or vascular endothelium NO-mediated vessel relaxation, etc. In addition, the direct/field tissue stimulation and/or extracellular field potential recording can be conducted together or separately from the bio-sensing and tissue tension studies, which enables many new experimental protocols.

    The bio-sensors, stimulation and/or recording electrodes are embedded as part of the tissue holding assembly, providing consistency and convenience in the experimental protocols without the need for external positioner. While the optional stimulation Pt. electrodes or recording metal micro electrodes are embedded on tissue holders, bio-sensors are installed on the sensor-, electrical- and drug-enabled tissue Measure & Position Bracket MP-20SED that also allows additional positioning of the bio-sensor. These biosensors come in two basic shapes in relation to the tissues being held: straight or L-shaped, as shown in the following diagrams. To include bio-sensor in your system order, please add “–SED” at the end of the system part number, and then specify what model of biosensor is needed.

    3. i-DAQChart Software Provides Tissue/Organ Bath Data Acquisition and Analyses

    i-DAQChart data acquisition and analysis software is an integral part of i-DAQ 4/8 or i-DAQTower-16, the portable, connect-and-play, function-rich data acquisition systems. It features real-time recording, effective on-line data smoothing/filtering one-line marking and commenting, automated cyclic measurements, data comparison, transformations, curve fittings and statistics analyses. The integrated i-DAQChart software contains special functions suited for the study and analyses of tissue/organ bath experiments, recorded on i-DAQ or BRAMDA-400/800. Among the important functions are the curve fitting, dose response curve and EC50/IC50, etc.

    i-DAQChart is available on two supported hardware: option #1: BRAM-4B Quad Bridge Amplifier plus i-DAQ 4/8 (4- or 8-ch) or i-DAQTower-16 (16-ch); option #2: BRAMDA-400/800 Quad/Octo Sensor Digital Recorder/Analyzer. As a third option, you can also use your existing software and data acquisition system.

    In this option, the portable, versatile data acquisition systems, i-DAQ4, i-DAQ8 or i-DAQTower16 with USB 2.0 is added as needed in the 4-, 8- or 16-channel EZ-Bath-4/8/16TBD configuration, which include 1x, 2x, or 4x BRAM-4B low noise quad bridge amplifiers with BNC outputs for i-DAQ or other data acquisition system. NO, O2 or glucose biosensor outputs from i-POTS Biosensor Potentiostat can be recorded and analyzed with i-DAQ as well.

    Option #2: Integrated 4- or 8-ch BRAMDA-400/800 Digital Sensor Record System w/ i-DAQChart Software

    In this option, i-FOT Force Transducers are plugged directly to the integrated system BRAMDA-400/800 with UBS port. In either options, all you need is a computer to start efficient, consistent experiments, recording and data analyses.



    Option #3: Using other data acquisition systems, bridge amplifiers or transducers

    If you’re using or already familiar with another data acquisition software and system, such as PowerLab or MP-150 Acknowledge, our BRAM-4B Bridge Amp outputs are compatible with these systems. Alternatively, our transducers can be adapted by re-wiring the DIN8 connector or using an adapting cable for these systems or other bridge amplifiers. If, however, you would like to use your transducers in the lab, our MP20 Measure and Position Bracket can be modified to accommodate your needs at minimal or no extra charge.
